It could be a number of things. Did you do the following when
setting up the source system:
1) Maintain the logical source system in BD54
2) Check to see that the user ID is the same for the connection
in both R/3 and BW, and the user has SAP_ALL & SAP_NEW
authority
3) Check that the R/3 and BW systems are set to "Modifiable"
4) Make sure that client is allowed to make change in both
client-dependent and independent objects in both systems
5) Check the RFC destination and ensure that the user can logon to target environment
6) Create the source system via the Administrator Workbench -> Source systems

Is the error you are getting say "Error is source system" ONLY, or are you getting any additional information? Sometimes it says "DataSource must be replicated" after attempting a dataload, for instance. However, if you are getting the "Error in source system" message after following the above steps to create the source system, then check the following:

1) RFC destination
2) Partner profiles. Ensure that they are active (i.e. on the classification tab check that "Partn. Status" = A and "Partner class" = BIW
3) Check that partner profile port numbers match up (i.e. we21)

In the future, avoid deleting the source system (after it is working) if something should ever happen and you think this would be a wise idea.
